Bremelanotide
PT-141, also known as Bremelanotide, is a synthetic peptide that works through the central nervous system to enhance sexual desire. Unlike PDE5 inhibitors that act on blood flow, PT-141 targets melanocortin receptors in the brain to address the neurological component of arousal.

PT-141 activates melanocortin-4 receptors (MC4R) in the hypothalamus, triggering signaling cascades that influence sexual arousal and desire. This central mechanism of action differentiates it from peripherally acting agents.
Common side effects include mild nausea, flushing, and headache, which typically diminish with subsequent doses. PT-141 should not be used more than once in a 24-hour period. Patients with uncontrolled hypertension should consult their provider.
